RESUMO

The stability analysis of underground caverns, tunnels, and boreholes is of great significance to underground engineering. In order to study the stress change of the coal around a hole and the evolution law of fractures during the coal seam drilling process, the discrete element simulation of the coal seam drilling process was carried out by using the particle flow code (PFC2D). The results show that during the drilling process, under the influence of confining pressure and drilling disturbance, the coal stress field around the hole and the development of fractures around the hole have the characteristics of zoning and dynamic evolution. In the axial direction of the borehole, it is divided into front and rear areas, and in the vertical axial direction, it is divided into the drilling disturbance zone and the confining pressure main control zone. During the drilling process, the direction of the maximum principal stress in the front zone gradually changes from the vertical hole axis to the direction parallel to the hole axis, and tension fractures are mainly developed along the drilling direction. In the rear zone, the principal stress direction tends to be stable and the principal stress value undergoes dynamic changes, and a large number of vertical hole axis tension fractures are developed. The drilling disturbance zone appears near the hole wall and has an important influence on the stability of the hole wall, while the confining pressure main control zone determines the antireflection effect around the hole and the influence radius of the hole. This work helps the understanding of the damage range and failure characteristics of the surrounding rock during the drilling process and has great significance for the guidance of drilling design.

Calciphylaxis is a rare disease characterized histologically by microvessel calcification and microthrombosis, with high mortality and no proven therapy. Here, we reported a severe uremic calciphylaxis patient with progressive skin ischemia, large areas of painful malodorous ulcers, and mummified legs. Because of the worsening symptoms and signs refractory to conventional therapies, treatment with human amnion-derived mesenchymal stem cells (hAMSCs) was approved. Preclinical release inspections of hAMSCs, efficacy, and safety assessment, including cytokine secretory ability, immunocompetence, tumorigenicity, and genetics analysis in vitro, were introduced. We further performed acute and long-term hAMSC toxicity evaluations in C57BL/6 mice and rats, abnormal immune response tests in C57BL/6 mice, and tumorigenicity tests in neonatal Balbc-nu nude mice. After the preclinical research, the patient was treated with hAMSCs by intravenous and local intramuscular injection and external supernatant application to the ulcers. When followed up to 15 months, the blood-based markers of bone and mineral metabolism improved, with skin soft tissue regeneration and a more favorable profile of peripheral blood mononuclear cells. Skin biopsy after 1-month treatment showed vascular regeneration with mature noncalcified vessels within the dermis, and 20 months later, the re-epithelialization restored the integrity of the damaged site. No infusion or local treatment-related adverse events occurred. Thus, this novel long-term intravenous combined with local treatment with hAMSCs warrants further investigation as a potential regenerative treatment for uremic calciphylaxis due to effects of inhibiting vascular calcification, stimulating angiogenesis and myogenesis, anti-inflammatory and immune modulation, multidifferentiation, re-epithelialization, and restoration of integrity.

Quantitative characterization of the pore structure and gas seepage characteristics of loaded coal is of great significance to the study of high-efficiency gas drainage in coal seams. Aiming at the problem of imperfect characterizations of coal seepage characteristics based on nuclear magnetic resonance (NMR), a calculation method for the pore permeability of coal with different pore diameters is proposed. The pore structure and seepage characteristics of coal have been quantitatively studied using a nuclear magnetic resonance (NMR) system. The results show that with increasing external load, the proportion of the pore volume of the coal sample in the range of 0.01-0.52 µm gradually decreases, while that in the range of 5.11-352.97 µm increases. In this process, the porosity increases from 0.9967 to 1.0103%, the connectivity increases from 0.1718 to 0.2391, and the permeability increases from 2.64 × 10-6 to 8.20 × 10-6 µm2. The calculation of the coal sample connectivity and permeability using the improved NMR permeability component proves that 94.37-352.97 µm pores are the main channel of fluid flow. When the axial pressure increases, the coal body permeability in the aperture range of 94.37-352.97 µm rapidly increases. The improved permeability component calculation model can better reflect the variation law of pore permeability of the loaded coal body.

Long-term peritoneal dialysis is accompanied by functional and histopathological alterations in the peritoneal membrane. In the long process of peritoneal dialysis, high-glucose peritoneal dialysis solution (HGPDS) will aggravate the peritoneal fibrosis, leading to decreased effectiveness of peritoneal dialysis and ultrafiltration failure. In this study, we found that the coincidence of elevated TGF-ß1 expression, autophagy, apoptosis and fibrosis in peritoneal membrane from patients with peritoneal dialysis. The peritoneal membranes from patients were performed with immunocytochemistry and transmission electron microscopy. Human peritoneal mesothelial cells were treated with 1.5%, 2.5% and 4.25% HGPDS for 24 hrs; Human peritoneal mesothelial cells pre-treated with TGF-ß1 (10 ng/ml) or transfected with siRNA Beclin1 were treated with 4.25% HGPDS or vehicle for 24 hrs. We further detected the production of TGF-ß1, activation of TGF-ß1/Smad2/3 signalling, induction of autophagy, EMT, fibrosis and apoptosis. We also explored whether autophagy inhibition by siRNA targeting Beclin 1 reduces EMT, fibrosis and apoptosis in human peritoneal mesothelial cells. HGPDS increased TGF-ß1 production, activated TGF-ß1/Smad2/3 signalling and induced autophagy, fibrosis and apoptosis hallmarks in human peritoneal mesothelial cells; HGPDS-induced Beclin 1-dependent autophagy in human peritoneal mesothelial cells; Autophagy inhibition by siRNA Beclin 1 reduced EMT, fibrosis and apoptosis in human peritoneal mesothelial cells. Taken all together, these studies are expected to open a new avenue in the understanding of peritoneal fibrosis, which may guide us to explore the compounds targeting autophagy and achieve the therapeutic improvement of PD.

BACKGROUND: Glutathione S-transferases (GSTs) are a family of multifunctional enzymes that are involved in the metabolism of many xenobiotics, including a wide range of environmental carcinogens. While the null genotypes in GSTM1 and GSTT1 have been implicated in tumorigenesis, it remains inconsistent and inconclusive. Herein, we aimed to assess the possible associations of the GSTM1 and GSTT1 null genotype in cancer risks. METHODS: A meta-analysis based on 506 case-control studies was performed. Odds ratios (OR) with corresponding 95% confidence intervals (CIs) were used to assess the association. RESULTS: The null genotypes of GSTM1 and GSTT1 polymorphisms were associated with a significantly increased risk in cancer (for GSTM1: OR = 1.17; 95%CI = 1.14-1.21; for GSTT1: OR = 1.16; 95%CI = 1.11-1.21, respectively). When the analysis was performed based on their smoking history, the risk associated of GSTM1 null and GSTT1 null genotypes with cancer is further increased (for GSTM1: OR = 2.66; 95%CI = 2.19-3.24; for GSTT1: OR = 2.46; 95%CI = 1.83-3.32, respectively). CONCLUSIONS: These findings indicate that GSTM1 and GSTT1 polymorphisms may play critical roles in the development of cancer, especially in smokers.

RESUMO

A dual-frequency liquid crystal based optical switch with orthogonally photo-patterned alignments is designed and fabricated. The cell gap is theoretically optimized for high switching performance. The measured extinction ratio of first diffraction order is over 20 dB with a low electric field of 3 V/µm. The switch On-Off time are measured to be 350 µs and 600 µs, respectively, both of which have reached submillisecond scale. Moreover, the switch is polarization independent, which has been predicted theoretically and further proved experimentally. This design is suitable in wide applications requiring fast response and polarizer free properties.

RESUMO

A diffraction grating is proposed by periodically defining the liquid-crystal director distribution to form alternate parallel aligned and twist nematic regions in a cell placed between two crossed polarizers. Based on the combined phase and amplitude modulation, both 1D and 2D tunable gratings are demonstrated. Low voltage ON/OFF switching of 1st order diffracted light with extinction ratio over 80 is achieved within a small voltage interval of 0.15 Vrms. Unique four-state feature of the cell is obtained and their applications in optical logic devices are discussed.
